Low Km = HIGH substrate affinity (Km is the [S] at ½Vmax). A low Km read as low affinity is exactly backwards.
Competitive inhibitors raise apparent Km but leave Vmax unchanged; non-competitive lower Vmax. Don’t swap them.

1 OD260 = 50 µg/mL for dsDNA, but ~40 for RNA and ~33 for ssDNA/oligos. Using 50 for all over-estimates RNA/ssDNA.
PCR
Wallace Tm = 4(G+C)+2(A+T) is valid only for SHORT oligos (<14 nt); long templates need nearest-neighbour methods.
